考马斯亮蓝染色

试剂： 1、Methanoh：acetic acid solution Combine 900 ml （500 ml of methanol and 400 ml of H2O) and 100 ml of glacial acetic acid. 2、Prepare the staining solution by dissolving 0.25 g of Coomassie Brilliant Blue R-250 per 100 ml of methanoli：acetic acid solution. 滤过through a Whatman No. 1 filter

步骤： 1、Immerse the gel in at least 5 volumes of staining solution ，至少缓慢摇动4 hours at room temperature.

2、Remove the stain and save it for future use. Destain the gel by soaking it in the methanol:acetic acid solution without the dye on a slowly rocking platform for 4-8 hours, 其间更换3-4次

3、After destaining, store the gels in H2O in a sealed plastic bag. 可将凝胶存放在含20%甘油的水中。

银染

试剂： 1、1%乙酸

2、显影液：2.5%的碳酸钠和0.02%甲醛的水溶液，新鲜配置 3、30%乙醇

4、固定液：Ethanoh：glacial acetic acid:H2O （30:10:60) 5、硝酸银溶液：现配现用，将20% AgNO3稀释成0.1% 步骤：

1、 将凝胶泡在至少5倍体积的固定液中，室温下4-12h

2、 去掉固定液，将胶泡在至少5倍体积的30%乙醇中，室温30min,shaking，并重复1次（1h）

3、 弃去乙醇，加入10倍体积的去离子水，室温10min，重复1次(20min) 4、 弃去水，加入5倍体积硝酸银溶液，室温30min

5、 弃去硝酸银溶液，去离子水漂洗凝胶2面，各20s(不可让胶脱水)

6、 加5倍体积显影液，数分钟内出现蛋白质的染色条带，至合适对比度，1%乙酸终止数分钟，再用去离子水清洗数次，每次10min
